    Skin penetration and conversion

    Panthenol penetrates the stratum corneum efficiently due to its favorable molecular weight (205 Da) and amphiphilic structure. Once inside keratinocytes, it is enzymatically oxidized to pantothenic acid (vitamin B5), the biologically active form.

    Coenzyme A synthesis

    Pantothenic acid is the precursor to coenzyme A (CoA), which is essential for lipid synthesis, fatty acid metabolism, and energy production in skin cells. Increased local CoA availability supports the synthesis of barrier lipids (ceramides, cholesterol, free fatty acids) in the stratum corneum.

Panthenol improves stratum corneum hydration by acting as a humectant and by stimulating lipid synthesis in the intercellular matrix. In controlled studies, 2% panthenol reduced TEWL by 20-25% over 4 weeks. Dexpanthenol (the D-isomer of panthenol) is the pharmacologically active form and is used in post-surgical wound care (Bepanthen) with extensive clinical evidence for skin barrier restoration.
