TB-500 is a synthetic analog of a naturally occurring region of Thymosin β4 (Tβ4) — a 43-amino-acid G-actin sequestering protein expressed in virtually every mammalian cell. The TB-500 fragment retains the actin-binding active site responsible for the most-studied downstream effects of Tβ4: cytoskeletal reorganization, enhanced cellular migration, accelerated re-epithelialization, and upregulation of VEGF-driven angiogenesis. Preclinical and early-clinical research with the full Tβ4 protein has explored corneal wound healing, epidermolysis bullosa, chronic pressure ulcers, and post-MI cardiac repair. The synthetic TB-500 fragment is used in research protocols exploring similar mechanisms at lower cost. Because of its longer biological half-life versus many other regenerative peptides, TB-500 is typically dosed less frequently (every 3–7 days in rodent models) rather than daily.
