Thymulin (formerly known as Facteur Thymique Sérique or FTS) is a nine-amino-acid peptide uniquely requiring a zinc ion for biological activity. It is the only thymic hormone with a fully characterized structure and a known metal cofactor requirement. Secreted by thymic epithelial cells, thymulin binds to high-affinity receptors on T-cell progenitors and promotes their differentiation into mature, immunocompetent T-cells. Circulating thymulin levels decline markedly with age — mirroring thymic involution — and are further suppressed by zinc deficiency, chronic stress, and viral infections. This age-related decline has positioned thymulin as a biomarker of immune aging and as a candidate intervention for immunosenescence. In preclinical models, thymulin administration has restored T-cell function in aged and thymectomized animals, reduced inflammatory cytokines in autoimmune models, and shown neuroprotective effects in demyelination paradigms. Human data is limited to small cohort studies and zinc-repletion trials.
