Selank is a synthetic peptide analog of the endogenous immunomodulatory peptide tuftsin (threonyl-lysyl-prolyl-arginine), developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. While its anxiolytic applications are the most commonly discussed use case, Selank's mechanism of action — modulation of GABAergic, serotonergic, and BDNF pathways — also supports cognitive enhancement, particularly under conditions of stress-related cognitive impairment, information overload, and sustained mental demand.

This protocol addresses cognitive enhancement specifically, building on but distinct from the anxiolytic protocol.

Mechanism overview: why Selank enhances cognition

Selank's cognitive effects are not a secondary consequence of anxiety reduction — though reduced anxiety certainly improves cognitive function. The peptide has independent procognitive mechanisms.

BDNF upregulation: Selank increases brain-derived neurotrophic factor expression, particularly in the hippocampus and prefrontal cortex. BDNF is the primary neurotrophin supporting long-term potentiation (the cellular basis of learning), synaptic plasticity, and memory consolidation. Higher BDNF levels correlate with better memory performance, faster learning, and greater cognitive resilience under stress.

GABA-A receptor modulation: Selank enhances GABA-A receptor sensitivity without producing sedation at therapeutic doses. This modulation reduces the excitotoxic "noise" from excessive glutamatergic activity — particularly relevant during stress, when elevated cortisol drives glutamate release that impairs prefrontal cortex function. The result is cleaner signal processing and improved working memory.

Serotonergic modulation: Selank influences serotonin metabolism, affecting the balance between serotonin synthesis and degradation. Serotonergic tone in the prefrontal cortex supports sustained attention, cognitive flexibility, and impulse control.

Enkephalin metabolism: Selank inhibits enkephalinase, the enzyme that degrades endogenous enkephalins. The resulting increase in enkephalin activity contributes to both anxiolysis and modulation of dopaminergic reward circuits involved in motivation and task engagement.

Protocol design

Primary protocol: Selank monotherapy

Compound: Selank, intranasal solution (typically 0.15% concentration = 150 mcg per spray)

Dose: 250-500 mcg per administration (2-3 sprays per nostril, depending on concentration)

Frequency: 2-3 times daily

Timing:

  • Morning dose (primary): 250-500 mcg within 30 minutes of waking. This dose establishes the baseline GABA/BDNF modulation for the day's cognitive demands.
  • Midday dose (maintenance): 250 mcg at midday (approximately 4-6 hours after morning dose). This sustains the cognitive support through the afternoon cognitive dip.
  • Early evening dose (optional): 250 mcg in the early evening (approximately 4-6 hours after midday dose) if sustained cognitive demand extends into the evening. Omit if not needed — this dose should be taken by 6-7 PM to avoid any potential interference with sleep onset in sensitive individuals.

Duration: 2-4 weeks per cycle. Russian clinical protocols typically run 14 days. Many practitioners extend to 21-28 days for cognitive applications where sustained BDNF elevation is the goal.

Intranasal administration technique

Proper technique determines whether the peptide reaches the olfactory epithelium (direct CNS access) or drains into the throat (systemic absorption with reduced CNS bioavailability).

Step 1: Clear nasal passages gently before administration. If congested, use a saline rinse 10-15 minutes before dosing. Do not administer Selank into a congested nostril — the peptide will not reach the olfactory epithelium.

Step 2: Tilt the head slightly forward (chin toward chest) — not backward. Tilting forward directs the spray toward the olfactory epithelium at the roof of the nasal cavity. Tilting backward directs the spray toward the throat, reducing CNS absorption.

Step 3: Insert the spray nozzle just inside the nostril, angling slightly toward the outer wall of the nasal cavity (toward the ear on the same side). This targets the olfactory cleft.

Step 4: Administer the spray while inhaling gently through the nose. Do not sniff forcefully — aggressive inhalation pulls the solution past the olfactory epithelium and into the nasopharynx.

Step 5: Wait 30-60 seconds between nostrils to allow absorption. Avoid blowing the nose for at least 15 minutes after administration.

Step 6: If administering both Selank and Semax (see combination protocol below), stagger by 15-30 minutes. Administering both simultaneously saturates the nasal mucosa and reduces absorption of both peptides.

Combination protocol: Selank + Semax

The Selank-Semax combination is the most widely used peptide nootropic stack, targeting complementary cognitive domains.

Selank provides anxiolytic tone, GABA modulation, and stress-buffered cognitive performance. It excels at reducing the cognitive cost of stress and anxiety — maintaining working memory and executive function under pressure.

Semax provides BDNF upregulation (stronger than Selank for this specific effect), dopaminergic and serotonergic stimulation, and cognitive activation. It excels at enhancing focus, processing speed, and creative problem-solving.

Why Selank first: Establishing the GABAergic anxiolytic tone before adding Semax's activating effects prevents the "wired but anxious" state that some individuals experience with Semax alone. Selank creates the calm foundation; Semax adds the cognitive edge.

Why no evening Semax: Semax has dopaminergic and mildly stimulating properties. While most users report no sleep interference, administering Semax after 4-5 PM introduces unnecessary risk. Selank, with its GABA-enhancing profile, is less likely to disrupt sleep and can be taken later if evening cognitive demands require it.

Amidate variants

NA-Selank-Amidate and NA-Semax-Amidate are modified forms with an acetyl group on the N-terminus and an amide group on the C-terminus. These modifications increase resistance to enzymatic degradation, extending the effective duration of each dose and potentially increasing potency per microgram.

If using amidate variants, reduce the dose by approximately 30-50% compared to standard forms and assess response before matching the standard dosing. The extended half-life may also mean that twice-daily dosing is sufficient where the standard form requires three administrations.

During the off phase, the cognitive effects do not disappear immediately. Many users report that the cognitive benefits partially persist for 1-2 weeks into the off period before gradually returning to baseline.

Extended cycling for chronic cognitive support

For individuals using Selank as ongoing cognitive support (e.g., during a 3-6 month period of sustained professional or academic demand), a longer cycling pattern may be appropriate:

4 weeks on, 2 weeks off, repeated for the duration of the demand period. The 2-week off periods prevent receptor desensitization while maintaining a higher proportion of time in the active phase.

Signs that suggest a longer off-period is needed

Diminishing subjective cognitive benefit despite consistent dosing (receptor desensitization). Increased baseline anxiety during the off period relative to pre-treatment baseline (suggestive of GABAergic tolerance). Irritability or mood instability when transitioning from active to off phase.

If any of these occur, extend the off-period to 4 weeks and reduce the subsequent active phase to 14 days. If they persist, take a full 8-week break before resuming.

Stacking considerations beyond Semax

Compatible additions

Magnesium L-threonate (1,500-2,000 mg daily, providing approximately 144-192 mg elemental magnesium): Crosses the blood-brain barrier more effectively than other magnesium forms. Supports synaptic density and NMDA receptor function. Complements Selank's GABA modulation without conflict.

Lion's mane mushroom extract (500-1,000 mg daily): Promotes nerve growth factor (NGF) production, complementing Selank's BDNF effects. Different neurotrophin pathways — NGF primarily supports cholinergic neurons while BDNF supports glutamatergic/GABAergic circuits. No known interaction with Selank.

Omega-3 fatty acids (EPA/DHA, 2-3 g daily): Structural support for neuronal membranes and anti-neuroinflammatory effects. Broadly supportive of the neuroplasticity that Selank and Semax promote.

Creatine monohydrate (3-5 g daily): Supports brain energy metabolism. The brain consumes 20% of the body's energy despite being 2% of its mass. Creatine provides a phosphocreatine buffer for ATP regeneration during high cognitive demand.

Combinations to avoid

Benzodiazepines: Both Selank and benzodiazepines target the GABA-A receptor. Combining them may produce excessive GABAergic sedation and does not provide additive cognitive benefit. If a patient is on prescribed benzodiazepines, Selank use should be discussed with the prescribing physician.

High-dose caffeine (above 400 mg/day): Excessive caffeine antagonizes adenosine receptors and promotes cortisol release, partially counteracting Selank's anxiolytic mechanism. Moderate caffeine (100-200 mg) is compatible and commonly combined in practice.

Other GABAergic compounds (phenibut, gabapentin, pregabalin): Stacking multiple GABA-modulating compounds increases the risk of excessive sedation and GABAergic tolerance. Choose one GABAergic agent for the protocol.

Monitoring and assessment

Cognitive function is inherently subjective, making protocol assessment more challenging than measuring IGF-1 levels or body weight. Structured self-assessment improves the signal-to-noise ratio.

Keep a brief daily log during the first cycle rating three dimensions on a 1-10 scale: focus/concentration, working memory (ability to hold and manipulate information), and stress reactivity (how quickly you become mentally derailed by stressors). Rate at the same time daily (midday is most informative). Compare the first 3 days of the active phase (before full effect) to days 7-14 (steady state) and to the off-period baseline.

If no meaningful improvement is documented after a complete 28-day cycle with consistent dosing and proper administration technique, Selank cognitive enhancement may not be the right intervention for that individual. The response to nootropic peptides is variable — absence of response is a valid outcome that should prompt re-evaluation rather than dose escalation.
