Palmitoyl Tetrapeptide-3 (Pal-GQPR) is a synthetic lipopeptide consisting of a palmitic acid moiety conjugated to the tetrapeptide sequence Gly-Gln-Pro-Arg. It belongs to the class of immunomodulatory cosmetic peptides designed to reduce cutaneous inflammation by suppressing pro-inflammatory cytokine release from dermal immune cells, particularly macrophages and keratinocytes. In-vitro studies demonstrate that Pal-GQPR significantly reduces the secretion of interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) in stimulated cell culture models, two key cytokines driving erythema, edema, and sensitization in inflammatory skin conditions. The palmitoyl chain enhances lipophilicity and stratum corneum penetration, improving delivery of the active tetrapeptide to the dermal compartment. Pal-GQPR is most commonly incorporated into formulations targeting sensitive, rosacea-prone, and post-procedure skin — contexts where chronic low-grade inflammation drives persistent redness and barrier dysfunction. It is frequently combined with Palmitoyl Tetrapeptide-7 (a complement cascade inhibitor) in commercial products marketed under trade names like Haloxyl or as components of multi-peptide calming serums. Unlike corticosteroids, which suppress inflammation broadly but cause skin thinning with chronic use, Pal-GQPR's mechanism is targeted at specific cytokine pathways without documented atrophogenic effects. Clinical evidence remains limited to manufacturer-sponsored in-vitro assays and small-scale observational studies — no independent randomized controlled trials have been published. Nevertheless, the anti-inflammatory rationale is mechanistically sound, and the peptide is well-tolerated with no reported irritation or sensitization.
