ACTH 4-10 (Met-Glu-His-Phe-Arg-Trp-Gly) is a heptapeptide fragment derived from positions 4 through 10 of adrenocorticotropic hormone (ACTH), a 39-amino-acid pituitary hormone best known for stimulating cortisol release from the adrenal cortex. Crucially, ACTH 4-10 retains none of the parent hormone's adrenal-stimulating (steroidogenic) activity — this resides in the 1-24 fragment — while preserving the behavioral and neurotrophic effects mapped to the 4-10 core sequence. This dissociation between endocrine and cognitive effects was recognized in the 1960s by De Wied and colleagues, making ACTH 4-10 one of the earliest peptide nootropics studied systematically. Human studies conducted primarily in the 1970s through 1990s demonstrated that ACTH 4-10 and its analogs (Org 2766, Semax contains a related sequence) improve attention, visual memory, and motivation in both healthy volunteers and patients with cognitive deficits. The peptide enhances selective attention and accelerates information processing without the arousal or stress responses associated with full-length ACTH. While the clinical trial data is more extensive than for many peptide nootropics, the studies are generally small, older, and have not been replicated with modern trial design standards. ACTH 4-10 remains a research compound with no current regulatory approval.
