Leuphasyl (trade name for the pentapeptide Tyr-Ala-Gly-Phe-Leu) is a synthetic cosmetic peptide designed to target the presynaptic side of the neuromuscular junction. Its mechanism involves mimicking enkephalin-like activity to reduce acetylcholine release at the motor end plate — effectively dampening the muscle contractions responsible for dynamic expression wrinkles. Unlike Argireline (acetyl hexapeptide-8), which competes with native SNAP-25 in the SNARE complex assembly, Leuphasyl is proposed to act via an opioid-receptor-mediated mechanism on the presynaptic neuron, reducing calcium influx and neurotransmitter vesicle fusion through a distinct upstream pathway. The peptide was developed by Lipotec (now part of Lubrizol) and is typically used in combination with Argireline rather than as a standalone. Manufacturer data suggests that the combination of Leuphasyl and Argireline produces greater wrinkle-depth reduction than either peptide alone — claimed at approximately 47% reduction in wrinkle depth versus 24% for Argireline alone in a 28-day split-face study. However, these results come from manufacturer-sponsored studies with limited independent replication. Leuphasyl is formulated at concentrations around 5% in topical serums and creams. It is well-tolerated, non-irritating, and compatible with other cosmetic actives. As with all topical neuromuscular peptides, the degree of stratum corneum penetration remains the primary bottleneck for efficacy, and results are modest compared to injectable neurotoxins.
