Argireline (acetyl hexapeptide-8, formerly acetyl hexapeptide-3) is a synthetic hexapeptide patterned on the N-terminal of SNAP-25, a SNARE protein required for neurotransmitter vesicle fusion. Its mechanistic claim is to competitively interfere with the SNARE complex and dampen acetylcholine release at the neuromuscular junction — the same target as botulinum toxin, but through a topical, reversible, and mechanistically less complete mechanism. In practice, the effect size is modest. Typical formulations at 5-10% show roughly 10-20% reduction in expression-wrinkle depth in split-face studies — meaningful but substantially less than intradermal botulinum toxin (which can approach 50-70% reduction). Argireline's appeal is as a needle-free, reversible, maintenance option or for users avoiding injectables. Real-world use is often combined with Matrixyl 3000 (which targets collagen synthesis rather than neurotransmission) for a complementary effect on both dynamic and static wrinkles.
