Pancragen is a synthetic short-chain peptide belonging to the class of 'peptide bioregulators' developed primarily by Russian researchers led by Vladimir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology. These bioregulators are typically di-, tri-, or tetrapeptides isolated from, or modeled on, fragments derived from specific organs, and are proposed to exert tissue-selective 'regulatory' effects on gene expression and cell function. Pancragen is the synthetic peptide associated with pancreatic tissue in this framework. The theoretical model behind the bioregulators holds that these short peptides can enter cells, reach the nucleus, and interact with DNA regulatory regions to modulate the expression of tissue-specific genes — in Pancragen's case, genes proposed to support the exocrine and endocrine pancreas and glucose-regulating function. Proponents describe applications in age-related pancreatic decline and carbohydrate metabolism support. It is important to be candid about the evidence base. The bioregulator literature is dominated by a relatively small group of investigators, much of it published in Russian-language journals, with limited independent replication and few rigorous, large-scale, placebo-controlled human trials indexed in mainstream databases. Mechanistic claims about direct peptide-DNA interaction remain incompletely validated by the broader scientific community.
