Palmitoyl pentapeptide-4, marketed under the trade name Matrixyl by Sederma (now part of Croda International), is a lipopeptide consisting of five amino acids (Lys-Thr-Thr-Lys-Ser) conjugated to a palmitoyl (C16) fatty acid chain. Launched in 2000, it was the first in the Matrixyl family of cosmetic peptides and rapidly became one of the most commercially significant and scientifically studied anti-aging peptide ingredients in the skincare industry. The subsequent Matrixyl 3000 (palmitoyl tripeptide-1 + palmitoyl tetrapeptide-7) and Matrixyl Synthe'6 expanded the family, but the original palmitoyl pentapeptide-4 remains widely used. The peptide was designed to mimic a fragment of the C-terminal propeptide of type I procollagen — a signaling peptide naturally released during collagen maturation that feeds back to fibroblasts to stimulate further collagen synthesis. This biomimetic approach leverages the body's own regulatory feedback mechanism: when collagen is being actively produced and processed, the released propeptide fragments signal fibroblasts to continue production. By delivering a synthetic analog of this signal topically, palmitoyl pentapeptide-4 is intended to upregulate the entire extracellular matrix production cascade. In vitro studies have consistently demonstrated that palmitoyl pentapeptide-4 stimulates fibroblast production of collagens I, III, and IV, as well as fibronectin, glycosaminoglycans including hyaluronic acid, and other extracellular matrix components. The palmitoyl tail is critical for efficacy — it provides the lipophilicity needed to penetrate the stratum corneum (the skin's primary barrier) and enhances cellular uptake through membrane interaction. Without the fatty acid conjugation, the hydrophilic pentapeptide alone has negligible skin penetration. Multiple published clinical studies have evaluated the anti-wrinkle efficacy of palmitoyl pentapeptide-4. A double-blind, placebo-controlled trial by Robinson et al. (2005) demonstrated significant reduction in wrinkle depth and volume after 12 weeks of twice-daily application, with improvements comparable to those observed with retinol formulations but without the irritation, photosensitivity, or retinoid dermatitis that limit retinol compliance. Profilometric analyses in additional studies have confirmed reductions in wrinkle depth of 15–37% over 2–4 months of use, depending on concentration and formulation vehicle.
