Palmitoyl Tetrapeptide-7 (formerly Palmitoyl Tetrapeptide-3) is a short-chain lipopeptide consisting of four amino acids (Gly-Gln-Pro-Arg) conjugated to a palmitoyl fatty acid chain. The palmitoyl lipid tail enhances skin penetration through the stratum corneum, allowing the peptide to reach the dermal layer where it exerts anti-inflammatory effects. Its primary mechanism is suppression of interleukin-6 (IL-6) and related pro-inflammatory cytokines, reducing chronic low-grade inflammation (inflammaging) that accelerates collagen breakdown and skin aging. It also inhibits the secretion of glycation-related damage mediators. Most commonly found as half of the Matrixyl 3000 complex — paired with Palmitoyl Oligopeptide — where it provides the anti-inflammatory component while its partner stimulates collagen synthesis.
